RESUMEN

Preeclampsia (PE) is a common pregnancy complication with a high mortality rate. Abnormally activated endoplasmic reticulum stress (ERS) is believed to be responsible for the destruction of key placental cells-trophoblasts. Phenylbutyric acid (4-PBA), an ERS inhibitor, is involved in regulating the development of ERS-related diseases. At present, how 4-PBA affects trophoblasts and its mechanisms is still unclear. In this study, PE cell models were established by stimulating HTR-8/SVneo cells with hypoxia. To verify the underlying mechanisms of 4-PBA on PE, CCT020312, an activator of PERK, was also used. The results showed that 4-PBA restored hypoxia-induced trophoblast viability, inhibited HIF-1α protein expression, inflammation, and PERK/ATF-4/CHOP pathway. Hoechst 33342 staining and flow cytometry results confirmed that 4-PBA decreased hypoxia-induced apoptosis in trophoblasts. The results of the JC-1 analysis and apoptosis initiation enzyme activity assay also demonstrated that 4-PBA inhibited apoptosis related to the mitochondrial pathway. Furthermore, by detecting autophagy in trophoblasts, an increased number of autophagic vesicles, damaged mitochondria, enhanced dansylcadaverine fluorescence, enhanced levels of autophagy proteins Beclin-1, LC3II, and decreased p62 were seen in hypoxia-stimulated cells. These changes were reversed by 4-PBA. Furthermore, it was observed that CCT020312 reversed the effects of 4-PBA on the viability, apoptosis, and autophagosome number of hypoxia-induced trophoblasts. In summary, 4-PBA reduces autophagy and apoptosis via the PERK/ATF-4/CHOP pathway and mitochondrial pathway, thereby restoring the viability of hypoxic trophoblasts. These findings provide a solid evidence base for the use of 4-PBA in PE treatment and guide a new direction for improving the outcomes of patients with PE.

Spores are important as dispersal and survival propagules in fungi. In this study we investigated the variation in number, shape, size and germination mode of ascospores in Morchella galilaea, the only species of the genus Morchella known to fruit in the autumn. Based on the observation of five samples, we first discovered significant variation in the shape and size of ascospores in Morchella. One to sixteen ascospores were found in the asci. Ascospore size correlated negatively with ascospore number, but positively with ascus size, and ascus size was positively correlated with ascospore number. We noted that ascospores, both from fresh collections and dried specimens, germinated terminally or laterally either by extended germ tubes, or via the production of conidia that were formed directly from ascospores at one, two or multiple sites. The direct formation of conidia from ascospores takes place within asci or after ascospores are discharged. Using laser confocal microscopy, we recorded the number of nuclei in ascospores and in conidia produced from ascospores. In most ascospores of M. galilaea, several nuclei were observed, as is typical of species of Morchella. However, nuclear number varied from zero to around 20 in this species, and larger ascospores harbored more nuclei. One to six nuclei were present in the conidia. Nuclear migration from ascospores to conidia was observed. Conidia forming directly from ascospores has been observed in few species of Pezizomycetes; this is the first report of the phenomenon in Morchella species. Morphological and molecular data show that conidial formation from ascospores is not found in all the specimens of this species and, hence, is not an informative taxonomic character in M. galilaea. Our data suggest that conidia produced from ascospores and successive mitosis within the ascus may contribute to asci with more than eight spores. The absence of mitosis and/or nuclear degeneration, as well as cytokinesis defect, likely results in asci with fewer than eight ascospores. This study provides new insights into the poorly understood life cycle of Morchella species and more broadly improves knowledge of conidia formation and reproductive strategies in Pezizomycetes.

The degradation of macroplastics results in micro/nanoplastics (MNPs) in the natural environment, inducing high health risks worldwide. It remains challenging to characterize the accurate molecular structures of MNPs. Herein, we integrate 193 nm ultraviolet photodissociation (UVPD) with mass spectrometry to interrogate the molecular structures of poly(ethylene glycol) terephthalate and polyamide (PA) MNPs. The backbones of the MNP polymer can be efficiently dissociated by UVPD, producing rich types of fragment ions. Compared to high-energy collision dissociation (HCD), the structural informative fragment ions and corresponding sequence coverages obtained by UVPD were all improved 2.3 times on average, resulting in almost complete sequence coverage and precise structural interrogation of MNPs. We successfully determine the backbone connectivity differences of MNP analogues PA6, PA66, and PA610 by improving the average sequence coverage from 26.8% by HCD to 89.4% by UVPD. Our results highlight the potential of UVPD in characterizing and discriminating backbone connectivity and chain end structures of different types of MNPs.

The orange subfamily (Aurantioideae) contains several Citrus species cultivated worldwide, such as sweet orange and lemon. The origin of Citrus species has long been debated and less is known about the Aurantioideae. Here, we compiled the genome sequences of 314 accessions, de novo assembled the genomes of 12 species and constructed a graph-based pangenome for Aurantioideae. Our analysis indicates that the ancient Indian Plate is the ancestral area for Citrus-related genera and that South Central China is the primary center of origin of the Citrus genus. We found substantial variations in the sequence and expression of the PH4 gene in Citrus relative to Citrus-related genera. Gene editing and biochemical experiments demonstrate a central role for PH4 in the accumulation of citric acid in citrus fruits. This study provides insights into the origin and evolution of the orange subfamily and a regulatory mechanism underpinning the evolution of fruit taste.

RESUMEN

A new species, Primula luquanensis Z.K.Wu and Wei Zhou sp. nov. (Primulaceae) is described and illustrated from Yunnan Province, China. It is morphologically assigned to P. sect Aleuritia based on its dwarf and hairless habit and coverage by farina on both sides of the leaf blade and scape. This new species is similar to P. nutantiflora and P. yunnanensis, but it is easily distinguished by its stolons, solitary bract, bell-shaped corolla and monomorphic floral form. The new species also has a substantially reduced corolla tube, presenting a unique floral form in a genus where heterostyly typically prevails.

BACKGROUND AND AIMS: Species of the genus Buddleja in Asia are mainly distributed in the Sino-Himalayan region and form a challenging taxonomic group, with extensive hybridization and polyploidization. A phylogenetic approach to unravelling the history of reticulation in this lineage will deepen our understanding of the speciation in biodiversity hotspots. METHODS: For this study, we obtained 80 accessions representing all the species in the Asian Buddleja clade, and the ploidy level of each taxon was determined by flow cytometry analyses. Whole plastid genomes, nuclear ribosomal DNA, single nucleotide polymorphisms and a large number of low-copy nuclear genes assembled from genome skimming data were used to investigate the reticulate evolutionary history of Asian Buddleja. Complex cytonuclear conflicts were detected through a comparison of plastid and species trees. Gene tree incongruence was also analysed to detect any reticulate events in the history of this lineage. KEY RESULTS: Six hybridization events were detected, which are able to explain the cytonuclear conflict in Asian Buddleja. Furthermore, PhyloNet analysis combining species ploidy data indicated several allopolyploid speciation events. A strongly supported species tree inferred from a large number of low-copy nuclear genes not only corrected some earlier misinterpretations, but also indicated that there are many Asian Buddleja species that have been lumped mistakenly. Divergent time estimation shows two periods of rapid diversification (8-10 and 0-3 Mya) in the Asian Buddleja clade, which might coincide with the final uplift of the Hengduan Mountains and Quaternary climate fluctuations, respectively. CONCLUSIONS: This study presents a well-supported phylogenetic backbone for the Asian Buddleja species, elucidates their complex and reticulate evolutionary history and suggests that tectonic activity, climate fluctuations, polyploidization and hybridization together promoted the diversification of this lineage.

Maintaining the protein high-order structures and interactions during the transition from aqueous solution to gas phase is essential to the structural analysis of native mass spectrometry (nMS). Herein, we systematically interrogate the effects of charge state and crown ether (CE) complexation on the gas-phase native-like protein structure by integrating nMS with 193 nm ultraviolet photodissociation (UVPD). The alterations of photofragmentation yields of protein residues and the charge site distribution of fragment ions reveal the specific sites and sequence regions where charge and CE take effect. Our results exhibit the CE complexation on protonated residues can largely alleviate the structure disruption induced by the intramolecular solvation of charged side chains. The influences of CE complexation and positive charge on gas-phase protein structure exhibit generally opposite trends because the CE microsolvation avoids the hydrogen-bonding formation between the charged side chains with backbone carbonyls. Thus, CE complexation leads to a more stable and native-like protein structure in the gas phase.

Unreactive C-H bond activation is a new horizon for frustrated Lewis pair (FLP) chemistry. This study provides a systematic assessment of the catalytic reactivity of recently reported intra-molecular FLPs on the activation of typical inert C-H bonds, including 1-methylpyrrole, methane, benzyl, propylene, and benzene, in terms of density functional theory (DFT) calculations. The reactivity of FLPs is evaluated according to the calculated reaction thermodynamic and energy barriers of C-H bond activation processes in the framework of concerted C-H activation mechanisms. As for 1-methylpyrrole, 14 types of N-B-based and 15 types of P-B-based FLPs are proposed to be active. Although none of the evaluated FLPs are able to catalyze the C-H activation of methane, benzyl, or propylene, four types of N-B-based FLPs are suggested to be capable of catalyzing the activation of benzene. Moreover, the influence of the strength of Lewis acid (LA) and Lewis base (LB), and the differences between the influences of LA and LB on the catalytic reactivity of FLPs, are also discussed briefly. This systematic assessment of the catalytic activity of FLPs should provide valuable guidelines to aid the development of efficient FLP-based metal-free catalysts for C-H bond activation.

This new species belongs to Vacciniumsect.Calcicolus and is most similar to V.brachyandrum, but differs in its branches hanging down, much smaller leaf blades, shorter inflorescences and pedicels, non-glandular tomentellate or densely pubescent inflorescence rachis and pedicels, densely white-pubescent hypanthium and pilose filaments.

In order to systematically evaluate the clinical efficacy and safety of misoprostol versus oxytocin in the prevention of postpartum hemorrhage, this paper provides evidence-based reference for clinical medication, computerized retrieval of Chinese biomedical literature database (CBM), PubMed, Embase, Cochrane Library, and clinical trials. The retrieval period is from the establishment of each database to October 1, 2021. Published randomized controlled trials (RCTS) are included in this study. The literature is screened and evaluated according to inclusion and exclusion criteria, and meta-analysis is performed using RevMan 5.3 software. A total of 13 RCTS are included, with a total of 24754 parturients. The meta-analysis shows the average blood loss (SMD = 0.10, 95% CI (-0.11, 0.32), P=0.35), the time of the third stage of labor (SMD = 0, 95% CI (-0.07, 0.08), P=0.95), and blood transfusion rate (RR = 0.80, 95% CI (0.63, 1.02), P=0.07). However, the incidences of shivering (RR = 2.61, 95% CI (1.79, 0.81), P < 0.00001) and vomiting (RR = 2.78, 95% CI (1.85, 4.18), P < 0.00001) are significantly higher than those in oxytocin group. The effect of misoprostol on preventing postpartum hemorrhage is similar to that of oxytocin, but the incidence of adverse reactions is high, and the occurrence of adverse reactions should be closely watched in the use process. Due to the limitations of the included studies, multicenter, large-sample, and high-quality RCTS are still needed in the future to further verify this conclusion.

Complex wrinkled microstructures are ubiquitous in natural systems and living bodies. Although homogeneous wrinkles in film-substrate bilayers have been extensively investigated in the past 2 decades, tailoring heterogeneous wrinkles by a facile method is still a challenge. Here, we report on the controllable heterogeneous wrinkles in template-modulated thickness-gradient metal films sputter-deposited on polydimethylsiloxane substrates. It is found that the stress of the gradient film is strongly position-dependent and the wrinkles are always restricted in thinner film regions. The morphological characteristic and formation mechanism of the heterogeneous wrinkles are analyzed and discussed in detail based on the stress theory. Ordered wrinkle arrays are achieved by adjusting the deposition time, copper grid period, template shape, and lifting height. The surface performances (e.g., the friction property) are well controlled by the wrinkle arrays. This work could promote better understanding of the spontaneously heterogeneous wrinkles in template-modulated gradient films and controllable fabrication of various wrinkle arrays by independently tuning film deposition conditions and template parameters.

Seed dispersal by frugivorous birds is a famous plant-animal interaction. However, we demonstrated that frugivorous birds can disperse seeds of Ligustrum lucidum, and its seed parasite weevils and the parasitic wasps from the weevils in this study. The cryptic tetrad of Ligustrum-disperser-weevil-wasp may be an overlooked phenomenon in seed dispersal syndrome.

Vacciniummotuoense (Ericaceae), a new species from Motuo County, Xizang Autonomous Region, China is described and illustrated. This new species belongs to Vacciniumsect.Calcicolus and is morphologically most similar to V.dunalianum, but differs in having yellowish-brown tomentose young branches, petioles and inflorescence rachis, leaf blades with 2-3(-4) pairs of secondary veins, usually all basal and with fine veins impressed adaxially and urceolate to spherical corollas.

Bio-desilication of fly ash containing a large amount of crystalline and amorphous silicate compound by silicate bacteria had the advantages of energy-saving, environment-friendly, and controllable cost. In this study, a bacteria strain with desilication ability on fly ash was isolated from the soil and identified as Bacillus amyloliquefaciens by 16sRNA and named ZGW-12. Through the bio-leaching treatment of fly ash by ZGW-12, it was found that the concentration of Si was 306.26 mg/L and the concentration of Al3+ was 0.5 mg/L in the pulp. The mechanism of bio-leaching using ZGW-12 was investigated by XRD, HPLC, SEM, and FTIR. The organic acid and amino acid in the bacteria culture medium and the pulp were detected, and it was found that the content of the acidic metabolites in the pulp was much larger than that of the bacteria culture medium. The ore sample particles adsorbed a large number of bacteria cells and took place obvious corrosion. The surface of the ore sample undergone hydroxylation reaction, and the diffraction peak of the silicate crystal compound was unchanged. ZGW-12 exhibits good performance in retaining aluminum and extracting silicon aspect. AVAILABILITY OF DATA AND MATERIAL (DATA TRANSPARENCY): All data were true and valid.

Humans have dyed textiles and leather, colored food, and paint body parts using natural dyes throughout history. Natural dyes have suffered drastically due to recent socioeconomic changes and replacement with synthetic dye in the textile industry. Hence, it is urgent to study indigenous dye plants and dyeing craft in local communities to protect these resources' potential ecological, economic, and cultural values. In 11 Monpa villages of Mêdog County, South-east Tibet, China, we conducted field research to record the indigenous method of cloth dyeing using madder dye. An aqueous extract of the root of Rubia wallichiana is a traditional madder dye. In this study, we used traditional dye and ethyl alcohol extract of the residue of aqueous extract. Two fabrics were dyed with the extractions in the presence of one of the metallic mordants or biomordants. Pigment compounds from aqueous extraction of madder and ethyl alcohol extraction of recycled madder were evaluated using Phytochemical, UV-visible spectroscopy, and FTIR test. We carried out One-way ANOVA and Duncan's new multiple range method to analyze different dying approaches and conditions. The dyed fabrics were evaluated by indicators of color strength and fastness, including washing, rubbing, and perspiration. The findings revealed the potentiality of biomordants to improve the dyeing properties of madder. The dyeing properties of recycled madder were marginally better than traditional madder. The results revealed the feasibility of enhancing the dyeing property and reuse of the residue from madder dyeing. The improved dyeing and reuse of residue can improve local ecological, economic benefits, and cultural heritage while applying research findings for the subsequent commercialization of plant dyes.

Primula homogama F. H. Chen & C. M. Hu (Primulaceae) is endemic to the Emei Mountain of China. In this study, we characterized the complete chloroplast genome of P. homogama based on next-generation sequencing (NGS). The complete chloroplast genome of P. homogama was 154,677 bp in size with a typical quadripartite structure, containing a large single-copy (LSC) region of 85,299 bp and a small single-copy (SSC) region of 17,816 bp. These two regions were separated by a pair of inverted repeat regions (IRs), each of 25,781 bp. A total of 130 functional genes were encoded, consisted of 86 protein-coding genes (PCG), 36 tRNA genes, and eight ribosomal RNA (rRNA) genes.

BACKGROUND: Home garden is identified as a kind of small-scale land-use system which is used to manage and cultivate useful plants by local people, and home gardens can provide various plant products and services. Investigating home gardens was regarded as an effective way to understand the biodiversity-related local knowledge and culture of native people in Ethnobiology and Ethnoecology. Home garden is important in less developed and remote areas. The grand canyon of Yarlung Tsangpo is designed as one of the biodiversity hotspots of China, and it is one of the most remote regions of China, because of the rough traffic conditions. The aim of the present study is to collect, record, and document the plants and their local knowledge and functions in the local home gardens, then attempt to answer the question: "why local people selected these plants?" MATERIAL AND METHODS: The study area was in Beibeng Township of Motuo County in the grand canyon of Yarlung Tsangpo. Observation and semi-structure interviews with informed consent were used to collect data in field study. All information was collected and organized, then documented based on "ethno-species" as a fundamental unit. All of the information of local use and knowledge were organized as the list of "use-report" for quantitative analysis, and the local uses of plants were merged into 14 use categories. Frequency of citation (FC), relative frequency of citation (RFC), cultural importance index (CI), and cultural value index (CV) were used in quantitative analysis. Besides, the Jaccard Index was used to compare the similarity in plant species selection among different communities. RESULTS: A total of 78 home gardens in the 9 communities of Beibeng Township were visited, and 196 ethno-species were collected. These ethno-species were identified into 188 Botanical taxa. A total of 87 home garden owners as informants were interviewed in the present study, and they provided 625 use-reports to us. The top 5 important plants were Su-lan-tsao (Dendrobium nobile), Sa-ga (Zingiber officinale), Soe-lu (Capsicum annuum), Snying-pa (Citrus medica), and Kham-pu (Prunus persica), according to the quantitative analysis. The most citied use-category was "vegetable," followed by "ornamental plant," "medicine," and "fruit." The altitude might be the most important impact factor of the plant diversity and composition of home gardens, and the traffic conditions, local terrain, also impact the plant diversity and composition of home gardens. CONCLUSION: In remote areas such as the Yarlung Tsangpo Grand Canyon, the plants in home gardens are important sources of plant products such as foods, herbal medicines, and fibers to support daily lives. The local home gardens in Tsang-la communities had high diversity of plants, and these plants provided many functions and services to support daily lives of local people. Local plant knowledge, including the features, life forms, habits, habitats, and use values of plants, were the summary of the understanding of local people to their surrounding plant worlds. Local people selected appropriate plants to cultivate and manage in their home gardens under the guidance of the local plant knowledge. That is the answer to the question "why local people selected these plants?"

BACKGROUND: Due to their relative isolation, the previous studies of Monpa plant use were only conducted in north-east India. In October 2013, Mêdog County was no longer remote, thanks to completion of a highway into the county. This study of plant species used by the Monpa had three research objectives. These were (i) to identify and record local names and uses of plants in Mêdog County, (ii) to assess which of these were uses of endemic or near-endemic species within this part of the Indo-Burma biodiversity hotspot, and (iii) to assess how plant uses reflect socio-economic change in Mêdog County? METHODS: Field surveys were conducted in 12 villages of four townships in Mêdog County, Tibet, China. Two field visits were made. The first field trip was in November 2017 and the second field trip was in May 2018. We interviewed 64 key informants between 21 and 84 years old. Most of them were the village leaders and other local people who are knowledgeable about plants. After transect walks with knowledgeable local people, we used free listing, key informant interviews, and semi-structured interviews during the field work. Plants traditionally used by the Monpa were documented. Utilization frequency was used to assess the significance of each species, and the Cultural Importance index was used to estimate the cultural significance of the species in common. We also used the informant consensus factor (FIC) to determine the homogeneity of the informants' knowledge of medicinal plants. RESULTS: One hundred ninety-four plant species belonging to 82 families and 158 genera were recorded and collected. One hundred twenty-two species, primarily fruits, were food plants. Forty-five species were used as traditional medicines. This included highly valued species collected in alpine areas (Paris polyphylla) and brought to villages in Mêdog, which are at a lower altitude (between 728 and 1759 m a.s.l). Seven edible plant species were also used as herbal medicines. We also recorded 39 species used for other purposes in Monpa daily life. These included nine species that were used to make agricultural tools, five species for dyes and mordants, four species for timber, three species for fuelwood, four species for religious ritual use, three species for washing, two species for incense, two species for thatching, two species for fiber (rope and paper), two "calendar plants" were used to indicate seasons for agricultural purposes, two fish poison plant species, and one species were used as a tobacco substitute. Based on taxonomic insights and from studies elsewhere, we suggested that fiber species were under-reported (c. 14 species were used vs. one species reported used). Even though these plant species are rich and diverse, the use of endemic or near-endemic species was rarely recorded in previous studies. These species included Arenga micrantha (used for starch), Hornstedtia tibetica (fruits), Castanopsis clarkei (edible nuts) and Gnetum pendulum (edible nuts), Ophiorrhiza medogensis (vegetables), Derris scabricaulis (fish poison), Radermachera yunnanensis (agricultural tools), Litsea tibetana (seed oil), Dendrocalamus tibeticus (wine strainers and implements for administering medicine), Zanthoxylum motuoense (spices), Cinnamomum contractum (tobacco substitutes), Morus wittiorum (medicines), and Garcinia nujiangensis (funeral rituals). Despite the absence of roads until 2013 and the impression of "isolation," Monpa knowledge of plant use reflects three categories of change. Firstly, oral histories of plants used in Bhutan were also encountered by Monpa people after their migration from Bhutan to south-eastern Tibet. Secondly, a "slow change" through centuries of exchange of knowledge (for example of Chinese and Tibetan medical systems), seeds of introduced crops (finger millet (indigenous to Africa), maize (from Meso-America)), and experimentation and use of introduced medicinal plants (such as Datura stramonium, which originates from North America). Thirdly, "fast change" over the past decade. This is reflected in changes in traditional architecture and in rising commercial trade in selected plant resources such as Dendrobium orchid stems and Paris polyphylla rhizomes which are in demand in China's Traditional Chinese Medicine (TCM) markets). CONCLUSIONS: Monpa people in the south-eastern Tibet have detailed knowledge of the diverse plant resources. But that traditional knowledge is now faced with a crisis because of the modern socio-economic change. In addition, Monpa knowledge of plants reflects slower changes in knowledge as well. For example, Monpa ethnomedicine has been influenced by traditional Tibetan and Chinese medicine over a longer period in time. Overall, this study provides a deeper understanding of the Monpa peoples' knowledge on wild plants, including endemic and near-endemic species whose uses have not been previously recorded. Several of these narrowly distributed species, such as the fish poison Derris scabricaulis, could be the focus of further studies. Some wild edible plants may also have interesting dietary constituents which need in-depth studies. These detailed studies could enable the Monpa people to benefit from the use of their traditional plant-derived culture and therefore support the biodiversity conservation.

RESUMEN

Bulbophyllum reflexipetalum, a new species from Motuo County, Southeast Xizang, China, is described and illustrated here. This new species belongs to Bulbophyllum sect. Umbellata Bentham & J. D. Hooker, and it is morphologically similar to B. umbellatum Lindley, B. guttulatum (J. D. Hooker) N. P. Balakrishnan and B. salweenensis X.H. Jin, but is distinguished from them by having reflexed petals, base of dorsal sepal with 1 dentate on each side, lip with significantly revolute margin, adaxially with dark brown spots or patches and one longitudinal groove.

A L. plantarum, CAU 3823, which can degrade 40% of biogenic amines (BAs) content in Chinese rice wine (CRW) at the end of post-fermentation, was selected and characterized in this work. It would be an optimal choice to add 106 cfu/mL of selected strain into the fermentation broth to decrease the BAs while keeping the character and quality of CRW. Nine amine oxidases were identified from the strain and separated using Sephadex column followed by LC-MS/MS analysis. The purified amine oxidase mixture showed a high monoamine oxidase activity of 19.8 U/mg, and more than 40% of BAs could be degraded. The biochemical characters of the amine oxidases were also studied. This work seeks to provide a better solution to degrade BAs in CRW prior to keeping the character and quality of CRW and a better understanding of the degradability of the strain to the BAs.
